G protein-coupled receptors (GPCRs) represent the key cell membrane receptors involved in all physiological and pathophysiological processes. Their importance is illustrated by the fact that they represent the targets of ~40% of the currently marketed drugs and a double recognition by Nobel Prizes awarded in 1994 and 2013 to scientific works related to GPCRs. Since their discovery our understanding of GPCRs has significantly evolved with the emergence of new concepts that will certainly help to understand their implication in human diseases and the design of safer and more efficient therapeutics. I will give a brief sight on GPCRs and the major advances regarding their pharmacology and signalling using the biophysical fluorescence and luminescence resonance energy transfer technologies.
